Experimental design. To reveal DAC-resistant mechanism, parental TF-1 and MDS-L cell lines were selected and their drug-resistant cell lines were developed. Parental and DAC-resistant cells were cultured in RPMI-1640 medium with or without drug treatment. The internal standard is composed of three cell lines (parental MDS-L, TF-1, U937) which were cultured in SILAC medium (13C6-Lys, 13C6-Arg) and equally mixed before nuclear isolation and acid extraction. Cells were collected and underwent nuclear isolation and acid extraction to achieve crude histone mixture. The light and heavy extracts were mixed equally followed by propionylation, trypsinization, LC–MS/MS and data analysis
